Up-regulation of the vitamin D receptor in response to 1,25-dihydroxyvitamin D3 results from ligand-induced stabilization.

چکیده
Several studies have shown that the 1,25-dihydroxyvitamin D3 (1,25-(OH)2D3) receptor protein levels increase in response to 1,25-(OH)2D3. We have studied the mechanism of this regulation in both mouse fibroblasts and rat intestinal epithelial cells. Cell extracts and total RNA were prepared at varying times after addition of 1,25-(OH)2D3. The 1,25-(OH)2D3 receptor protein levels, measured using an immunoradiometric assay, rose significantly 2-3 h posttreatment and had risen 3-fold at 8 h. Concurrently, the 1,25-(OH)2D3 receptor mRNA content, measured using a ribonuclease protection assay, was not altered by 1,25-(OH)2D3 during this time. In cycloheximide-blocked cells, the administration of 1,25-(OH)2D3 markedly reduced the degradation rate of previously formed receptor. The 1,25-(OH)2D3 receptor protein half-life was determined as 4 h in the absence of 1,25-(OH)2D3 and increased to at least 8 h in the presence of 1,25-(OH)2D3. We also measured the 1,25-(OH)2D3 receptor mRNA levels in the duodena and kidney of vitamin D-deficient rats after a single 150-pmol injection of 1,25-(OH)2D3. Again, we found that 1,25-(OH)2D3 receptor mRNA levels were not changed in these tissues after 1,25-(OH)2D3 treatment. Therefore, the elevation of the 1,25-(OH)2D3 receptor protein following 1,25-(OH)2D3 administration is apparently the result of increased receptor protein lifetime and not increased transcription.

1, 25 Dihydroxyvitamin D3 Protects the Heart Against Pressure Overload-induced Hypertrophy without Affecting SIRT1 mRNA Level
Background and Aims: There has been scant information concerning antihypertrophic effects of vitamin D specifically on its cellular and molecular mechanisms. Sirtuin 1 (SIRT1) is regarded as a key deacetylase enzyme in cardiomyocytes which applies potential cardioprotective effects by functional regulation of different proteins. This study aimed to evaluate the effects of 1, 25-dihydroxyvitamin...
متن کاملPhosphorylation of the 1,25-dihydroxyvitamin D3 receptor. A primary event in 1,25-dihydroxyvitamin D3 action.
1,25-Dihydroxyvitamin D3 is believed to mediate the regulation of calcium metabolism by a steroid hormone receptor mechanism. We now demonstrate that phosphorylation of the 1,25-dihydroxyvitamin D3 receptor occurs rapidly in embryonic chick duodenal organ culture in response to 1,25-dihydroxyvitamin D3. Phosphorylation of the receptor is strongly induced within 1 h by 1,25-dihydroxyvitamin D3 a...
متن کاملThe Association of Vitamin D Receptor Gene BsmI Polymorphism with Multiple Sclerosis in Iranian Patients
Background & Aims: 1,25-dihydroxyvitamin D3 (1,25 (OH)2 D3), the biologically active form of vitamin D, exerts an immunosuppressive effect through binding to its specific nuclear receptor. The present case-control study was done to examine the possible association of BsmI polymorphism in vitamin D receptor gene (VDR gene) with severity of multiple sclerosis (MS). Methods: 267 Iranian patients w...
متن کاملRegulation of the vitamin D receptor and cornifin beta expression in vaginal epithelium of the rats through vitamin D3.
The aim of the present study was to determine the respective role of 1,25-dihydroxyvitamin D3 on vaginal epithelium and 1,25-dihydroxyvitamin D3 receptor expression in ovariectomized rats and vitamin D3 treated rats. Bilateral ovariectomies were performed in 20 mature, non-pregnant Wistar female rats. All the animals were divided into 2 groups consisting of 10 rats each. Group I served as contr...
متن کاملConformational change and enhanced stabilization of the vitamin D receptor by the 1,25-dihydroxyvitamin D3 analog KH1060.
The 1,25-dihydroxyvitamin D3 [1,25-(OH)2vitamin D3] analog KH1060 exerts very potent effects on cell proliferation and cell differentiation via the vitamin D receptor (VDR). However, the activities of KH1060 are not associated with an increased affinity for the VDR. We now show that increased stabilization of the VDR-KH1060 complex could be an explanation for its high potencies. VDR half-life s...
